Rate-limiting and tightly regulated step, in the proximal renal tubule: up-regulated by PTH and hypophosphataemia, suppressed by FGF-23 and by calcitriol itself.

Inactivation arm; induced by calcitriol. Loss-of-function causes idiopathic infantile hypercalcaemia.
