Nucleoside diphosphate kinase (NDPK/NME)

What it does, reaction by reaction

Deoxyribonucleotide Formation (Ribonucleotide Reductase pathway and thymidylate synthesis) Nucleotide Metabolism · Cytosol

step 3 Irreversible/directional

dADP, dGDP, dCDP, or dUDP → corresponding dNTP

Converts dADP dGDP dCDP dUDP into dATP dGTP dCTP dUTP

Notes

NDPK catalyzes dNDP + ATP (or another NTP) ⇌ dNTP + ADP. This near-equilibrium phosphotransfer uses a phosphohistidine intermediate and supplies dATP, dGTP, dCTP, and dUTP from the RNR products.

step 10 Irreversible/directional

dTDP → dTTP

Converts dTDP into dTTP

Notes

NDPK catalyzes dTDP + ATP (or another NTP) ⇌ dTTP + ADP. dTTP then participates in DNA polymerization and also serves as an allosteric specificity effector for RNR and an inhibitor of dCMP deaminase.

Pyrimidine De Novo Synthesis (through to UMP, then CTP/dTMP) Nucleotide Metabolism · Cytosol (one mitochondrial step)

step 8 Irreversible/directional

UDP → UTP

Converts UDP into UTP

Notes

Nucleoside diphosphate kinase (NDPK/NME) catalyzes UDP + ATP (or another NTP) ⇌ UTP + ADP (or the corresponding NDP). A phosphohistidine enzyme intermediate mediates phosphoryl transfer; the reaction is near equilibrium.

step 11 Irreversible/directional

dUDP → dUTP → dUMP

Converts dUDP into dUTP

Notes

NDPK can phosphorylate dUDP + ATP ⇌ dUTP + ADP, and dUTPase then catalyzes dUTP + H2O → dUMP + PPi. dUTPase is essential because it both supplies dUMP and keeps dUTP low enough to prevent uracil incorporation into DNA.
