Hormone-sensitive lipase (HSL; LIPE)
Cofactors used
Clinical / pharmacological. Activated by PKA (glucagon/epinephrine), inhibited by insulin
What it does, reaction by reaction
Triacylglycerol Synthesis and Lipolysis Lipid Metabolism · Cytosol / ER
DAG + H2O → monoacylglycerol (MAG) + free fatty acid
Converts Diacylglycerol H2O into Monoacylglycerol (MAG) Fatty acid
› Notes
Hormone-sensitive lipase (HSL; LIPE) hydrolyzes DAG most efficiently; it also has TAG and cholesteryl-ester hydrolase activity. PKA phosphorylation of HSL and perilipin promotes access to the lipid droplet.
Showing all 1 reactions.
What accelerates and inhibits it
Regulation is pathway-specific, so each context is listed separately
Ketogenesis
Listed there as: Hormone-sensitive lipase/ATGL in adipose (upstream fuel supply)
Catecholamines, glucagon in humans, natriuretic peptides
Insulin; phosphodiesterase-mediated cAMP degradation
Low insulin and counterregulatory hormones increase nonesterified-fatty-acid delivery to liver
Triacylglycerol Synthesis and Lipolysis
Listed there as: Hormone-sensitive lipase (HSL)
PKA phosphorylation; phosphorylation of perilipin; catecholamines/natriuretic peptides
Dephosphorylation; insulin; phosphodiesterase 3B lowering cAMP
Epinephrine and norepinephrine activate; insulin strongly inhibits; glucagon has a modest/direct role in human adipocytes compared with catecholamines
